چکیده

Radioautographic and radiochemical techniques were used to establish the presence of replicating DNA in the chloroplasts of Acetabularia mediterranea. These techniques also demonstrated that these chloroplasts synthesize RNA. It was found that label from thymine was also incorporated into DNA and RNA in these chloroplasts. With the establishment of protein and nucleic acid synthesis in Acetabularia chloroplasts, it is clear that these chloroplasts carry out those metabolic processes which are most characteristic of autonomous cells.

Studies on Independent Synthesis of Cytoplasmic Ribonucleic Acids in Acetabularia mediterranea

1. The RNA content of anucleate and nucleate fragments of Acetabularia has been measured. It was found that there is a net synthesis of RNA in nucleate fragments. On the other hand, the RNA content of anucleate fragments did not change significantly after enucleation. 2. Anucleate fragments, however, can readily incorporate (14)C-labeled adenine, orotic acid, and carbon dioxide into their cytop...

Biosynthesis in Isolated Acetabularia Chloroplasts

The ability of chloroplasts isolated from Acetabulana mediterranea to synthesize the protein amino acids has been investigated. When this chloroplast isolate was presented with (14)CO(2) for periods of 6-8 hr, tracer was found in essentially all amino acid species of their hydrolyzed protein Phenylalanine labeling was not detected, probably due to technical problems, and hydroxyproline labeling...

Base composition and metabolism of various RNA fractions in Acetabularia mediterranea.

The fact that anucleate fragments of Acetabularia mediterranea are capable of regeneration and morphogenesis and of specific protein synthesis has been known for many years." 2 The information needed for these processes obviously comes from the nucleus and must be stored for a considerable time in the cytoplasm: this can be most readily explained in molecular terms by assuming that the nucleus ...

An Analysis of Cellular and Subcellular Systems Which Transform the Species Character of Acid Phosphatase in Acetabularia

Several species-specific molecular forms of acid phosphatase are known to exist in the unicellular green alga Acetabularia. In graft combinations between cells of Acetabularia mediterranea (med) and Acicularia Schenckii (acic) the expression of the med phosphatase is dominant over acic phosphatase. There is good evidence that in such grafts the preexisting acic phosphatase is converted on the m...

Chloroplast membranes of the green alga Acetabularia mediterranea. II. Topography of the chloroplast membrane

The localization of the chlorophyll-protein complexes inside the thylakoid membrane of Acetabularia mediterranea was determined by fractionating the chloroplast membrane with EDTA and Triton X-100, by using pronase treatment, and by labeling the surface-exposed proteins with 125I. The effects of the various treatments were established by electrophoresis of the solubilized membrane fractions and...
